Objectives

The aims of the course are to provide students with the methodological knowledges regarding the use of the immunohistochemical and molecular techniques in clinical practice and research. The acquired informations will allow the student to understand the variety of approaches that can be used to study the tissues and to prepare them to make the most appropriate methodological choices to solve an experimental or diagnostic issue and to critically analyze the experimental results.

Contents

The teaching module aims to provide the student with the methodological bases regarding the ancillary techniques used in a Pathology lab, with particular attention reserved to the immunohistochemical and molecular biology techniques applied to human tissues (in situ hybridization techniques), and to provide the knowledge tools for the correct choice of the technique able to respond to a clinical or research question.

- Principles of the immunohistochemical staining
- How to set up a histological sample for the immunohistochemical staining
- Types of antibodies and methods for the production of monoclonal and polyclonal antibodies
- Physico-chemical properties of the antigen-antibody binding: causes of the antigen unbinding and methods to optimize the use of the immunoreagents
- Antigen identification through immunodetection systems
- Antigen retrieval methods: procedures using heat and protein digestion
- Principal immunohistochemical staining methods
- Pitfalls and technical artifacts in immunohistochemistry
- Application of immunohistochemistry in human pathology
- Tissue Microarray (TMA) sample preparation
- Methods for automatic sample preparation and analysis
- Diagnostic roles of the molecular biology techniques (i.e., FISH, CISH and RNAscope methods) in human pathology
- Use of the cDNA and RNA microarray technology for the study and characterization of tumors

Verification of learning:

The course will be held through frontal lessons. The student preparation will be tested through oral questions with the final exam score expressed in thirtieths. The student must have learned the general principles of the immunohistochemical staining methods applied to the human tissues, and of the main molecular biology techniques.
